What do human studies actually show? 

In a four-week randomized trial, 128 adults experiencing low mood received placebo, 22 mg, or 28 mg per day of Affron®, a standardized saffron stigma extract. The 28 mg group showed significant improvements on the study's total mood-disturbance score compared to placebo. The 22 mg dose did not produce the same pattern, which is an important reminder that a botanical name without a dose tells only half the story (Kell et al., 2017). 

An eight-week trial of 56 healthy adults with subclinical low mood used 30 mg of standardized saffron daily. Researchers reported improvements in some mood and social-relationship outcomes, along with a measurable saffron metabolite in urine. Not every emotional measure changed, and the group was small (Jackson et al., 2021). 

A larger 2025 trial followed 202 adults with low mood for 12 weeks. Participants received 28 mg of Affron® or placebo. The saffron group had a modestly greater improvement on the study's main mood scale, with an effect size of 0.39. A placebo response was also present, which is common in mood research and keeps the result in perspective (Lopresti et al., 2025). 

Not every trial has been positive. A separate 2025 randomized study of 51 adults used 30 mg of standardized saffron for six weeks and found no significant advantage on its primary composite of mood, anxiety, and fatigue measures. One secondary mental-health measure favored saffron, but secondary findings do not erase a negative primary result (Amadieu et al., 2025). 

The newest relevant trial, published in 2026, enrolled 86 women aged 50 to 70 who reported low mood and poor sleep. After 12 weeks, 28 mg of Affron® was associated with modest improvements in the primary mood score, self-esteem, and sleep-related daytime impairment (Lopresti and Smith, 2026). 

The evidence from these studies supports a careful conclusion that standardized saffron around 28 to 30 mg per day may support emotional well-being and upbeat mood for some adults, particularly when used for several weeks, but it is not a guaranteed mood transformation and it depends from one person to another. 

How quickly should you expect to notice a difference? 

The trials lasted four to 12 weeks. That makes “feel everything in one cup” a poor expectation.
